Unit 8 Objectives- Analysis and Characterization of Nucleic Acids and Proteins

  1. Describe how restriction enzyme sites are mapped on DNA.
  2. Compare and contrast Northern, Southern, and Western blot hybridzation methods including the target, probe and purpose.
  3. List and describe the three methods used to tranfer the fragments separated by electrophoresis to a solid medium.
  4. State the purpose of baking or UV cross-linking the denatured DNA after tranferring to the membrane.
  5. State the purpose of the prehibridization step in the Southern blot procedure.
  6. Calculate the melting temperature of a given sequence of dsDNA.
  7. Describe the basic method and uses of the following hybridization technologies: dot blot, slot blot, reverse dot blot, macroarray, microarry, high density oligonucleotide array and microelectronic array techniques.
  8. Be able to define or describe the terms listed in the "Unit 8 Glossary of Terms".
